Job Description

This internship opportunity is for undergraduates. It is a full-time paid position (40 hours per week) located at our facility in Agoura Hills, California. The internship will begin mid-late June and continue for 10 weeks.

The Process Development team is seeking a Summer Intern to gain experience in Cell Biology and automation. As an intern, you will be trained and mentored by some of the brightest minds in the field and exposed to different aspects of the biotech industry. You will acquire valuable hands-on skills and experience while contributing to our science and pipeline.

You will also have the opportunity to take on a project to support Lentiviral Vector (LVV) potency assay development. LVVs are widely used in gene therapy partly due to their capability to stably integrate in both dividing and nondividing cells. Transgene expression and potency assessment of clinical-grade LVV lots are required by regulatory agencies to support lot release of this critical drug substance for marketing application. The LVV bioassay will be a cell-based, quantitative assay that detects both transgene expression and biological activities. The procedures to measure transgene expression from transduced primary T cells by both flow cytometry and ddPCR have been developed.

For this project, you will work on developing an in-vitro potency assay using well characterized cell lines with relative potency readout. Initial assessment on assay reproducibility, optimum time point, generating and qualifying standard reagents are expected. Further assay development and optimization may be performed for late-stage development with the methodology applicable for other T-mod programs. This work will have a direct impact on the advancement of A2’s autologous T-mod programs to pivotal development stage. You will be mentored by an experienced PhD and gain knowledge on all aspects of

potency assay development. Most importantly, our internships are designed to prepare and fuel passionate individuals to thrive as they continue their career paths in cell therapy field. The student will have the opportunity to interact with the whole Process Development Department with emphasis on the Analytics Team and report to The Analytics Team Lead, who is a Senior Process Development Engineer. Some of the outcomes of this work are expected to be implemented into A2 workflows of this assay and therefore are pivotal in advanced of A2 program pipelines.

Internship Responsibilities:

  • Responsibilities will include (but are not limited to):
  • Conduct assigned experiments and document experiment results in a timely manner
  • Gain knowledge and basic skills on the operation and data acquisition of lab equipment such as flow cytometer, plate reader etc.
  • Contribute to the experiment design, trouble shooting and data sharing within Process Development team
  • Meet regularly with mentors and attend Process Development group meeting.
  • Review published literature to acquire a broader understanding of A2’s science and your project.

Requirements

  • Currently enrolled in a full-time undergraduate program preferably at Junior year status (“Rising Senior”) at an accredited university / college in biology, biochemistry, biotechnology, bioengineering or a related discipline
  • Legally authorized to work in the United States without A2 sponsorship
  • Basic understanding of cell biology attained during academics
  • Ability to execute experiments, analyze and present data independently
  • Strong collaboration and communication skills
  • Self-motivated and well-organized with the ability to work independently and as part of a team
  • Scientific curiosity and good attention to detail
